At the end of protein induction, the form of a-SMA expression was determined by a Elisa kit(HB246-Ch, Shanghai hengyuan biological technology co., LTD) respectively. The induced proteins were solubilized using a lysis solution of 8 M urea. Ni-NTA gravity protein purification columns (Sangon, China) were chosen to purify the proteins. The efficiency of 100 mM, 200 mM, 300 mM, 400 mM, and 500 mM imidazole eluents was examined separately to determine the most efficient concentration of imidazole in the eluent. The purified proteins were dialyzed and concentrated, mixed with glycerol and stored at 80C.
